چکیده

Ethanol and β-galactosidase production from cheese whey may significantly contribute to minimise environmental problems while producing value from lowcost raw materials. In this work, the recombinant Saccharomyces cerevisiae NCYC869-A3/pVK1.1 flocculent strain expressing the lacA gene (coding for β-galactosidase) of Aspergillus niger under ADHI promoter and terminator was used. This strain shows high ethanol and β-galactosidase productivities when grown on lactose. Batch cultures were performed using SSlactose medium with 50 gL lactose in a 2-L bioreactor under aerobic and microaerophilic conditions. Temperature was maintained at 30 ° C and pH 4.0. In order to determine the effect of an electric field in the fermentation profile, titanium electrodes were placed inside the bioreactor and different electric field values (from 0.5 to 2 Vcm) were applied. For all experiments, β-galactosidase activity, biomass, protein, lactose, glucose, galactose and ethanol concentrations were measured. Finally, lag phase duration and specific growth rate were calculated. Significant changes in lag phase duration and biomass yield were found when using 2 Vcm. Results show that the electric field enhances the early stages of fermentation kinetics, thus indicating that its application may improve industrial fermentations’ productivity. The increase in electric field intensity led to plasmid instability thus decreasing β-galactosidase production.

FED-BATCH BIOREACTOR PROCESS WITH RECOMBINANT Saccharomyces cerevisiae GROWING ON CHEESE WHEY

Saccharomyces cerevisiae strain W303 was transformed with two yeast integrative plasmids containing Kluyveromyces lactis LAC4 and LAC12 genes that codify β-galactosidase and lactose permease respectively. The BLR030 recombinant strain was selected due to its growth and β-galactosidase production capacity. Different culture media based on deproteinized cheese whey (DCW) were tested and the best ...

Development of Stable Flocculent Saccharomyces cerevisiae Strain for Continuous Aspergillus niger β-Galactosidase Production

A flocculent Saccharomyces cerevisiae strain was engineered to stably secrete Aspergillus niger β-galactosidase in a continuous high-cell-density bioreactor. The δ-sequences from the yeast retrotransposon Ty1 were used as target sites for the integration of the β-galactosidase expression cassette. High-copy-number transformants were successfully obtained using the δ-integration system together ...

Isolation and Identification of Yeast Strains Capable of Producing Single Cell Protein from Whey in Co-Cultures with Saccharomyces cerevisiae

In this study, twenty-five whey samples collected from dairy industries in the city of Isfahan. The sampleswere cultured on malt extract broth (MEB) and yeast extract glucose chloramphenicol agar (YGCA) media.Eleven yeast strains (designated M1 to M11) were isolated from the culture. The strains were identified bytheir morphological and physiological properties. Betagalactosid...

In this study, immobilization of partially purified almond (Amygdalus communis) β-galactosidase on Con A layered calcium alginate-cellulose beads was investigated. Immobilized β-galactosidase retained 72% of theinitial activity after crosslinking by glutaraldehyde. Both soluble and immobilized enzyme exhibited the samepH and temperature optima at pH 5.5 and 50ºC, respectively. Howev...
